Comments: Characteristics: Transfected with reverse tetracycline-controlled transactivator (rtTA), a fusion between a mutated version of E.coli TetR and the activating domain of HSV-1 VP16., Characteristics: Unlike the parental cell line, this cell line maintains mtDNA at normal levels. This was achieved by introducing into the parental cell line a ligA gene fused to an excisable (FRT/FLPo) mitochondrial targeting sequence (MTS) from human ornithine transcarbamylase. The resulting cell line expresses two ligA variants: one without- and another with MTS. The latter is efficiently imported into mitochondria and therefore normal mtDNA copy number is restored in the transduced cells (Kerafast=ESA104).
