Comments: From the depositor: In 1998, we purchased from Invitrogen pSecTag2A and cut it with BglII and BamH1. The BglII/BamH1 fragment was replaced with a modified fragment containing 6 Histidines at the N-Terminus that was done by generating the modification by PCR, and cloning the PCR product back into BglII/BamH1. Sometime later, the above pSecTag2A-6HIS was digested with BamH1and Not1. The ATX (amino acids 52-863) insert was genereated by PCR, digested with BamH1/EcoR1 and by excising ATX from the original colony II cDNA library with EcoR1/Not1, which yields two pieces in two stages into the vector. Their is a stop codon before the C-Terminus Myc tag so it will not be expressed.
