Comments: IpaA cDNA was amplified from pEC14, supplied by M. Venketesan (Walter Reed Army Institute of Research, Washington, DC), using appropriate primers to introduce an NdeI site before the initiating ATG and an XhoI site downstream of the stop codon. The PCR product was transformed into TOPO pCRII (Invitrogen). The NdeI and XhoI fragment of IpaA cDNA was isolated from a pCRII clone containing the correct sequence and inserted into pET15b vector (Novogen) to generate pET15b/IpaA.
