Comments: Plasmid D588 (lys2::HIS3 ) was constructed in two steps. First, plasmid YDp-K was cut with BglII and XhoI and ligated, deleting a 2477 bp fragment inside the LYS2 gene. This plasmid was then cleaved with BglII and a 1.2 kb BamHI fragment from YDp-H containing HIS3 was inserted, creating D558.
