This was achieved by introducing into the parental cell line a ligA gene fused to an excisable (FRT/FLPo) mitochondrial targeting sequence (MTS) from human ornithine transcarbamylase. The resulting cell line expresses two ligA variants: one without- and another with MTS. The latter is efficiently imported into mitochondria and therefore normal mtDNA copy number is restored in the transduced cells (Kerafast=ESA104).
