This antibody reacts with human Sprouty-2 protein and is determining its presence in ELISA, immunoblotting and immunoprecipitation. Approximately 20 ul of antibody is used to immunoprecipitate hSpry2 protein from 1 mg of a HeLa cell lysate in 1 ml. Incubate the antibody for 2 h with the cell lysate. Immunoprecipitate the complex with Protein G agarose. Reactivity in other immunoassays has not yet been fully determined.

This affinity purified antibody was assayed by immunoblot and was found to be reactive at a 1:2,000 dilution. A 38 kDa band corresponding to human Sprouty-2 is observed. By immunoblotting against the reduced protein, the antibody identifies bands at 30 kDa and 18 kDa (zymogen and active MMP26). MMP26 does not appear to be produced by most normal quiescent cells, but the treatment of many cell types with the phorbol ester TPA or IL1 stimulates production of MMP26. Because of the low protein levels produced (pg/ml)concentration of cell culture media is often required to visualize the bands byimmunoblotting. Note: Higher antibody dilutions may be necessary for non-human samples. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.
