Comments: "Monoclonal antibody anti-zebrin II is secreted by a murine hybridoma cell line produced from a mouse immunized with a crude homogenate from the electric lateral line lobe (ELL) and cerebellum of the weakly electric fish Apteronotus leptorhynchus. Each of a panel of mice were immunized intraperitoneally with 5 mg of protein emulsified in Freund's complete adjuvant. Four more injections in incom-plete adjuvant were given at 3 week intervals. Finally the animals were boosted with ELL/cerebellum homogenate in saline. Three days after the final injection, the mice were bled, tested for anti-cerebellum/ELL titer by using a dot immunobinding assay and the mouse with the highest titer was selected for hyhridoma production. Technical details of the fusion, cell culture, and cloning were as used previously. Positive clones were selected for their immunostaining patterns on frontal sections of fixed gymnotiform brain. In all, 480 independent primary wells were screened from this fusion of which about 25% secreted anti-ELL/cerebellum antibodies. The anti-zebrin II specificity was cloned by limiting dilution. Because the antibody reveals a pattern of stripes in the rat cerebellum (see Results), we have named the antigen zebrin II."
