Comments: "For antibody production, an N-terminal portion of Nok (aa 27–280) was fused to glutathione S-transferase, expressed in yeast and purified using an ESP system (Stratagene). We used 1 mg of purified Nok-GST to immunize a rabbit (Cocalico Biologicals). Rabbit serum was subjected to several rounds of purification on affinity columns, prepared by conjugating Nok or GST to agarose beads using an AminoLink Plus Immobilization Kit (Pierce). In the final step, anti-Nok antibodies were eluted with 0.1 M glycine, pH 2.5, precipitated with saturated ammonium sulfate and re-dissolved in 1×PBS with 50% glycerol. We carried out western blotting according to standard procedures."
