  • RRID:Addgene_172356

http://www.addgene.org/172356

Genetic Insert: scbR
Vector Backbone Description: Backbone Size:5715; Vector Backbone:pSET152; Vector Types:Bacterial Expression, Synthetic Biology, Integrative into Streptomyces genome via serine integrase (like parent pSET152); Bacterial Resistance:Hygromycin
Defining Citation: PMID:29130169
Comments: Biarnes-Carrera, Marc, Chang Kwon Lee, Takuya Nihira, Rainer Breitling, and Eriko Takano. ‘Orthogonal Regulatory Circuits for Escherichia Coli Based on the γ-Butyrolactone System of Streptomyces Coelicolor’. ACS Synthetic Biology 7, no. 4 (2018): 1043–55. https://doi.org/10.1021/acssynbio.7b00425. pTE1062 is replicative in E. coli via pMB1 type origin, and carrying oriT (RP4) for delivery to Streptomyces by conjugation. In Streptomyces, pTE1062 is not replicative, and therefore selection is on integration into the genome mediated by phiC31 integrase.

Please note that Addgene's sequencing results identified a single nucleotide mismatch at bp#4947 when compared to the full sequence information provided by the depositing laboratory. This mismatch is not a concern for the function of the plasmid.

Vector Backbone Description: Vector Backbone:pBAD33 + pTrc99a; Vector Types:Bacterial Expression; Bacterial Resistance:Hygromycin
Defining Citation: PMID:31672972
Comments: pSiMPlh_N should be cotransformed with pSiMPlh_C (or vice versa) in E. Coli to get a reconstituted functional HPT thereby conferring resistance against hygromycin. mRuby3 gene was amplified from a construct bought by us from Addgene (ID: 74252). Please see the Sequences link for the individual plasmid sequences. The sample from Addgene will have both the plasmids pSiMPlh_N + pSiMPlh_C in it for the interested antibiotic resistance. Separating individual plasmid backbones will be possible via PCR. Agarose gel electrophoresis can be used to visualize the presence of both the plasmids. But, separating individual plasmids via gel electrophoresis, in our hands, always resulted in cross-contamination of the two backbones.
