  • RRID:Addgene_108254

http://www.addgene.org/108254

Vector Backbone Description: Vector Backbone:N/A; Vector Types:; Bacterial Resistance:Chloramphenicol
Defining Citation: PMID:30478458
Comments: The three new “Marionette” strains contain the Marionette cluster, which expresses 12 inducible transcription factors, from the genome. Plasmids pAJM.711, pAJM.712, pAJM.713, pAJM.714, pAJM.715, pAJM.717, pAJM.716, pAJM.718, pAJM.719, pAJM.1459, pAJM.721 are reporters for those 12 transcription factors. For examples: TetR is being expressed from the genome of the Marionette strains, and pAJM.717 has a promoter that is repressed by TetR (PTet*) driving YFP. pAJM.717 should not produce YFP, until the cell is exposed to anhydrotetracycline (aTc). Other plasmids from our laboratory encode both the reporter and the transcription factor, and should be used in a regular (non-Marionette) strain. For example: pAJM.011 expresses TetR and contains a PTet*-YFP reporter module. Please visit https://www.biorxiv.org/content/early/2018/04/10/285866 for bioRxiv preprint.

Vector Backbone Description: Vector Backbone:pSepT; Vector Types:Bacterial Expression; Bacterial Resistance:Chloramphenicol
Defining Citation: PMID:21868676
Comments: To prevent possible enzymatic dephosphorylation of O-phospho-L-serine (Sep) in vivo, the gene encoding phosphoserine phosphatase (serB), which is catalyzing the last step in serine biosynthesis, was deleted from Escherichia coli strains Top10 (Top10∆serB - Addgene #34928) and BL21 (BL21∆serB - Addgene #34929). These strains are required hosts when using this plasmid.

http://www.addgene.org/34879

Genetic Insert: SB100X transposase
Vector Backbone Description: Vector Backbone:pCMV; Vector Types:Mammalian Expression; Bacterial Resistance:Chloramphenicol
Defining Citation: PMID:19412179
Comments: The Sleeping Beauty transposon system consists of two components: the transposon vector and the transposase vector. The transposon vector (pT4/HB, Addgene #108352) contains binding sites flanking the GOI. And the transposase vector (pCMV(CAT)T7-SB100, Addgene #34879) encodes for the enzyme mediating excision and integration of the GOI.

Vector Backbone Description: Vector Backbone:pBbA5c; Vector Types:Bacterial Expression, Synthetic Biology; Bacterial Resistance:Chloramphenicol
Defining Citation: PMID:21952217
Comments: Please note that the R364 in Mevalonate Kinase (MK) may be susceptible to mutations. Although the original plasmid encoded R364C, Addgene's stock contains R364S. This mutation is not expected to impact overall function, however the exact effects are unknown.
