  • RRID:Addgene_220280

http://www.addgene.org/220280

Vector Backbone Description: Backbone Size:5256; Vector Backbone:pSpy0C4; Vector Types:Bacterial Expression; Bacterial Resistance:Chloramphenicol
Defining Citation: PMID:38911550
Comments: The multiple cloning site (EcoRI, XbaI, SpeI and PstI) contains the Plac_mrfp cassette. The mrfp reporter allows for red-white screening during sub-cloning in E. coli (DH5a). When the Plac_mrfp cassette is replaced with the insert of interest, colonies will appear white. Resource Information: The MCS featuring the Plac_mrfp cassette and the chloramphenicol resistance cassette were taken from pSpy1C. The pUC origin for replication in E. coli was taken from pUC19-mKate2∼ssrA. Up- and downstream homologous regions of sagB were amplified from the S. pyogenes SF370 genome. Linearise the plasmid with the restriction enzyme Alw44I before transformation in S. pyogenes to favour double crossover recombination of the plasmid into the sagB locus. Positive transformants will show loss of hemolysis on TSA blood agar plates. Please download the file linked below to obtain a well annotated map of the plasmid.

Species: Photinus pyralis
Genetic Insert: ffluc
Vector Backbone Description: Vector Backbone:pSpy1C; Vector Types:Bacterial Expression, Luciferase; Bacterial Resistance:Chloramphenicol
Defining Citation: PMID:38911550
Comments: This high-copy replicative shuttle plasmid features an erythromycin-inducible riboswitch driving expression of the Firefly luciferase in the presence of erythromycin. Luminescence can be detected by adding luciferin to the culture. See associated publication for more details. Resource Information: The erythromycin-inducible riboswitch was taken from pLZ12Km2-TA:PgyrA(Sag)-ermBL-ermB':A2I:ffluc-tt3, which was used in the following publication: Dynamics of diversified A-to-I editing in Streptococcus pyogenes is governed by changes in mRNA stability Thomas F. Wulff, Karin Hahnke, Anne-Laure Lécrivain, Katja Schmidt, Rina Ahmed-Begrich, Knut Finstermeier, Emmanuelle Charpentier bioRxiv 2023.09.19.555891; doi: https://doi.org/10.1101/2023.09.19.555891 The ffluc reporter gene was taken from the original pLZ12Km2-P23R:TA:ffluc plasmid (Plasmid #88900). • RRID:Addgene_220248

http://www.addgene.org/220248

Vector Backbone Description: Backbone Size:4917; Vector Backbone:pSpy1C; Vector Types:Bacterial Expression; Bacterial Resistance:Chloramphenicol
Resource Information: We took the rolling circle replicon pSH71 and the chloramphenicol resistance cassette from pIB166 (Plasmid #90189). The the Plac_mrfp cassette for the MCS is derived from pBS3Clux (Plasmid #55172). Please download the file linked below to obtain a well annotated map of the plasmid.
