Sometime later, the above pSecTag2A-6HIS was digested with BamH1and Not1. The ATX (amino acids 52-863) insert was genereated by PCR, digested with BamH1/EcoR1 and by excising ATX from the original colony II cDNA library with EcoR1/Not1, which yields two pieces in two stages into the vector.
