Comments: Genomic sequence subcloned by recombineering from a clone of RPCI-11 human genomic DNA library. The exact DNA sequence in this plasmid differs somewhat from the reference human genome sequence because the reference sequence is a composite sequence, i.e., the reference ID1 region sequence may not be derived from the RPCI-11 BAC clone utilized for this construct.
The ID1 intron was placed after G198, i.e., after about a third of the sequence encoding the active site of the cre protein. See https://doi.org/10.1073/pnas.2011448117 and https://doi.org/10.1093%2Fnar%2Fgkq384. Thus, without splicing, no catalytically active cre protein sequence is expected to be translated from this sequence.
