Vector Backbone Description: Backbone Marker:Malcolm Moore; Backbone Size:8326; Vector Backbone:pUltra-hot (Addgene Plasmid #24130); Vector Types:Mammalian Expression, Lentiviral, Multicistronic; Bacterial Resistance:Ampicillin
Comments: pUltra-hot-H2B-mTagBFP2 was cloned by Jorge L. Arias-Arias at Universidad de Costa Rica ([email protected]).
This vector allows the multicistronic expression of mCherry, H2B-mTagBFP2, and one gene of interest in mammalian cells. The construct contains two 2A peptides coding sequences and two cloning positions flanked by restriction sites:
Position 1 (5' XbaI – 3' BamHI): Occupied by the H2B-mTagBFP2 insert.
Position 2 (5' NheI – 3' EcoRI or SaII): Available for cloning one gene of interest.
The vector is suitable for transient transfection, but could also be used to produce lentiviral particles for stable expression in mammalian cells. For lentivirus packaging use the additional plasmids psPAX2 (Addgene Plasmid #12260) and pMD2.G (Addgene Plasmid #12259).

Species: Homo sapiens
Genetic Insert: TFAM Homology Arms with linker-mEGFP
Vector Backbone Description: Vector Backbone:pUC57; Vector Types:Mammalian Expression, CRISPR, Donor Template; Bacterial Resistance:Ampicillin
Comments: This plasmid has been used with locus-specific CRISPR/Cas9 to add a mEGFP tag to the C-terminus of human TFAM in WTC-11 human induced pluripotent stem cells by the Allen Institute for Cell Science. Linker (AA) sequence: GGGS. After protein tagging using this donor template plasmid and CRISPR/Cas9 reagents, transfected cells may exhibit varying intensity levels of fluorescence, likely due to editing precision. To obtain cells of uniform intensity levels, see our protocol for fluorescence-assisted cell sorting and subcloning of transfected cells (https://www.allencell.org/instructional-videos-and-tutorials-for-cell-methods.html). Further, we recommend PCR-based assays for identifying precisely edited clones as previously described (https://www.molbiolcell.org/doi/abs/10.1091/mbc.e17-03-0209). For more information on the entire plasmid collection, please see https://www.addgene.org/allen-institute-cell-science/ .
