http://www.addgene.org/20793

Species: Danio rerio
Genetic Insert: mir-206-1/mir-133b
Vector Backbone Description: Backbone Marker:Bartel Lab; Backbone Size:0; Vector Backbone:pBSII SK+ modified to introduce GFP and more cloning sites; Vector Types:zebrafish expression; Bacterial Resistance:Ampicillin
Defining Citation: PMID:19240133
Comments: For polycistronic mir-206-1/mir-133b, we were not able to identify the transcriptional start precisely by 5′-RACE due to the repetitive sequence in this region. Analyses of EST clones (BM259633, CF999541, CF348604, DR729161) mapped the approximate start of the mir-206-1/mir-133b primary transcript to 1.4 kb from the mir-206 hairpin. A 4.5-kb upstream fragment that includes ∼0.9 kb of the primary transcript was amplified from genomic DNA using primers 3 and 4 (5'-CTGCCCCATTTAGACTCTTTCTTGACG-3' and 5'-CCAGAAGAACCCAGACGAAACCCAC-3'). The promoter fragment was fused to a GFP reporter gene. The miRNA promoter fusion was flanked by I-SceI meganuclease recognition sites to increase the efficiency of transgenensis. Please note that Addgene's sequencing results show some differences from the published sequence. The depositing lab is aware of the differences.

Species: Danio rerio
Genetic Insert: mir-124
Vector Backbone Description: Backbone Marker:Bartel Lab; Backbone Size:0; Vector Backbone:pBSII SK+ modified to introduce GFP and more cloning sites; Vector Types:zebrafish expression; Bacterial Resistance:Ampicillin
Defining Citation: PMID:19240133
Comments: 5′-RACE (GenRacer kit, Invitrogen) identified a transcriptional start of the mir-124-5 primary transcript, which is located 1.1 kb from the mir-124-5 hairpin. A 5.6-kb fragment upstream of the mir-124-5 transcriptional start was amplified from genomic DNA using primers 1 and 2 (5'-TGCTTTGAGGCACGGTTATGGTCCC-3' and 5'-CTCGGGCAGCGGCTTTTTGGTCCTG-3'). The promoter fragment was fused to a GFP reporter gene. The miRNA promoter fusion was flanked by I-SceI meganuclease recognition sites to increase the efficiency of transgenensis.

http://www.addgene.org/24334

Species: Danio rerio
Genetic Insert: H2B
Vector Backbone Description: Backbone Size:3000; Vector Backbone:pBluescript; Vector Types:Worm Expression, Cre/Lox; Bacterial Resistance:Ampicillin
Defining Citation: PMID:19706417
Comments: This plasmid was constructed using 1.5 kb of the hsp70l promoter. The floxed mCherry-STOP and H2b_GFP fragments were PCR amplified individually and sequentially cloned downstream of the hsp70l promoter. The mCherry-STOP cassette serves as a heat-shock induction marker prevents read-through translation of the H2B-GFP fusion protein, which is only expressed in cells that have undergone a Cre mediated excision event. cryaa-Cerulean was inserted downstream of HSB-GFP in the reverse orientation. *H2B has D26G and V119I mutations. According to the depositor, GFP expression from this construct was always nuclear so there is no reason to suspect that the mutations affect the function (nuclear localization) of the H2B fragment.

Species: Danio rerio
Genetic Insert: fli1a intron 1
Vector Backbone Description: Backbone Size:2858; Vector Backbone:p5E; Vector Types:; Bacterial Resistance:Kanamycin
Defining Citation: PMID:23462469
Comments: "p5E" refers to 5' entry clones flanked by L4-R1 There are a few mismatches between depositor's sequence and Addgene's sequence. The changes should not affect plasmid function.

Proper citation: RRID:Addgene_26027 Copy

Species: Danio rerio
Genetic Insert: miR-126
Vector Backbone Description: Backbone Size:0; Vector Backbone:pME; Vector Types:Gateway Cloning; Bacterial Resistance:Kanamycin
Defining Citation: PMID:20364122
Comments: The insert includes genomic sequence flanking the mature miR-126 sequence on chromosome 8 (miRBase accession number MI0001979, Zv8). There is a C->T mutation in the miR126. The mutation is probably due to a SNP and should not affect plasmid function.

  • RRID:Addgene_26436

    This resource has 1+ mentions.

http://www.addgene.org/26436

Species: Danio rerio
Genetic Insert: flk1 promoter
Vector Backbone Description: Backbone Marker:Invitrogen; Backbone Size:3973; Vector Backbone:pCRII-TOPO; Vector Types:Topo cloning vector; Bacterial Resistance:Ampicillin
Defining Citation: PMID:16251212
Comments: From the paper: Approximately 6.5 kb of upstream sequence of the zebrafish flk1 gene (Liao et al., 1997; Thompson et al., 1998) was amplified from wild-type genomic DNA based on the sequence information previously deposited (from –6410 to –3 of the transcriptional start site, GenBank Accession Number AY045466) (Chan et al., 2002), and subcloned into the pCRII TOPO vector (Invitrogen). This fragment has been shown to drive endothelial-specific expression of the green-red coral fluorescent protein (G-RCFP) (Cross et al., 2003). The EcoRI cleaved fragment from this construct was placed 5′ upstream of the enhanced green fluorescent protein (EGFP) vector (Clontech) to drive endothelial specific expression of EGFP
