Comments: For maintenance of vector, any standard E.coli is OK, but to make plasmid preps, the plasmid must be propagated in EPI300 or similar strain that enables the arabinose-inducible copy control system. Induction of gene expression is with acetamide in Mycobacterium, but this doesnt work in E.coli. Some level of control over expression in E.coli can be gained via controlling the copy number of the plasmid with arabinose. See published paper for more details on construction and use of pMycoFos. Comments: pUS116 is an E.coli/Mycobacterium shuttle vector, with high copy number in E.coli and low copy number in Mycobacterium. The plasmid has unique NdeI, BamHI, PvuII, EcoRI, NheI sites for cloning, and genes cloned at this MCS can be expressed via acetamide induction in Mycobacterium, but this system doesnt work in E.coli. This plasmid was made by deletion of the recombineering genes from pJV53 (see van Kessel and Hatfull, 2007), to re-forma basic shuttle/expression vector. pUS116 is equivalent to vector pLAM12.
