Species: S. pyogenes, Z. mays
Genetic Insert: Cas9
Vector Backbone Description: Backbone Marker:Promega; Vector Backbone:pGEM-T easy; Vector Types:CRISPR, Gateway Cloning; Bacterial Resistance:Chloramphenicol and Ampicillin
Defining Citation: PMID:31523744
Comments: Also has aminoglycosyl phosphotransferase driven by 35S promoter (G418 resistance). We are not sure of the backbone -- it came from a plasmid that was made many many years ago. It aligns well with pGEM-T Easy (which a lot of our plasmids are based on) so if we had to guess, we would say the backbone is pGEM-T Easy.

Vector Backbone Description: Backbone Marker:Dirk Landgraf; Backbone Size:5056; Vector Backbone:pDML112; Vector Types:Yeast Expression, Synthetic Biology; Bacterial Resistance:Chloramphenicol and Ampicillin
Defining Citation: PMID:29056345
Comments: This is the empty basic yTRAP sensor plasmid. To sense aggregation of your favorite protein, clone the coding sequence into a pENTR clone, then perform LR reaction with pGAN147. The resulting expression clone should be linearized with NotI-HF enzyme prior to integration into the yeast genome. The sensor integrates into the HO locus, and can be selected for using nourseothricin.

Defining Citation: PMID:21107422
Comments: The AtAMT1;1 coding sequence with the att sites (attB1 and attB2) from the pDRf1-AtAMT1;1 (Loque et al 2007 Nature 446 195-198) was remobilized with both restriction enzymes SpeI and XhoI then subsequently cloned between SpeI and XhoI sites in the p6013-002 (Ludewig et al., 2002 J. Biol. Chem. 277, 13548-13555). The AtAMT1;1 coding sequence was then replaced by the Gateway DNA cassette after a BP reaction with pDONR 221 (GATEWAY technology, Invitrogen) to generate the p002-GW vector.
