Comments: pUS116 is an E.coli/Mycobacterium shuttle vector, with high copy number in E.coli and low copy number in Mycobacterium. The plasmid has unique NdeI, BamHI, PvuII, EcoRI, NheI sites for cloning, and genes cloned at this MCS can be expressed via acetamide induction in Mycobacterium, but this system doesnt work in E.coli. This plasmid was made by deletion of the recombineering genes from pJV53 (see van Kessel and Hatfull, 2007), to re-forma basic shuttle/expression vector. pUS116 is equivalent to vector pLAM12.
