There is a unique KpnI site immediately following the XhoI site at the 3' end of the cloned fragment. The NotI site and BglII sites are also usable for anyone wanting to reclone the fragment and the XhoI site is not usable but the KpnI site is.

  • RRID:Addgene_67525

http://www.addgene.org/67525

Species: Drosophila melanogaster
Genetic Insert: crumbs
Vector Backbone Description: Backbone Size:8904; Vector Backbone:pUAST; Vector Types:Insect Expression, Drosophila; Bacterial Resistance:Ampicillin
Comments: The plasmid contains part of the 5' cDNA of Crb and the 3' part that brings the transmembrane and the intracellular domains of the protein together. The BglII restriction site links the 5' and 3' regions of the cDNA, meaning that the majority of the extracellular part of the Crb protein is missing. The fragment also contains the 5' and 3' UTR regions. BglII restriction site was used to clone a GFP fragment. GFP is inserted between aa 96 and 2122 of crb. Crb contains K89E mutation which does not affect plasmid function.

Species: Drosophila melanogaster
Genetic Insert: Drosophila melanogaster Shaker cognate l
Vector Backbone Description: Backbone Size:3200; Vector Backbone:pBSc; Vector Types:Xenopus Oocyte expression; Bacterial Resistance:Ampicillin

  • RRID:Addgene_29327

http://www.addgene.org/29327

Species: Drosophila melanogaster
Genetic Insert: UAS-Stop-GFP:HSV-mTFP1:HA-mKO2:V5
Vector Backbone Description: Backbone Marker:Pfeiffer et al; Backbone Size:7831; Vector Backbone:pJFRC-MUH; Vector Types:Insect Expression; Bacterial Resistance:Ampicillin
Defining Citation: PMID:21297621
Comments: This plasmid contains mTFP1.0, and not EBFP2, as suggested by the linear plasmid map image file. Please note that Addgene's sequencing results identified a few discrepancies when compared to GenBank ID JF267350. The depositing laboratory states that these differences are not a concern for the function of the plasmid. The full plasmid sequence has been assembled and not entirely confirmed by sequencing. Additional discrepancies may exist. This plasmid was constructed using the following restriction enyzmes: Bgl II / Not I - loxP sites Not I / Xho I - EGFP-HSV Xho I / Kpn I - mTFP1.0-HA Kpn I / Xba I - mKO2-V5 NotI is not a unique cutter--it is present twice in the plasmid. The XbaI site has been confirmed by Addgene's sequencing results, but it not present in the full plasmid sequence.
