It consists of a 5' homology arm, CAG promoter, FRT-flanked neomycin resistance and stop cassette, LoxP-flanked EcoRV cloning site and stop cassette, PmeI cloning site, WPRE and bgh pA elements, and a 3' homology arm. A subtractive cDNA of interest (expressed in response to FLP but not Cre recombination) can be cloned into the EcoRV site. The intersectional cDNA of interest (expressed in response to both Cre and FLP recombination) can be cloned into the PmeI site. This vector lacks the sgRNA target sequence found in the px330_Rosa_sgRNA plasmid.
