RRID:Addgene_35195 Chloramphenicol and Ampicillin PMID:22848718 Generation of ϕC31 transgenic Drosophila. Gateway cloning cassette is flanked by gypsy insulator sequences to allow uniform transgene expression between insertion sites. Transgene selection using white gene. loxP site facilitates elimination of transgene markers via Cre recombinase-mediated excision with ZH-attP landing sites. Ampicillin resistant. Chloramphenicol resistant for amplification. Please see the associated publication for more information: Wang J-W, Beck ES, McCabe BD (2012) A Modular Toolset for Recombination Transgenesis and Neurogenetic Analysis of Drosophila. RRID:Addgene_35194 PCP-2x-yeGFP Synthetic Ampicillin PMID:21512033 This plasmid expresses a chimeric RNA-binding protein fused to GFP used to detect mRNAs that contain multiple copies of an RNA sequence (see Addgene plasmid #'s 35191, 35192 & 35196). See associated publication for detailed methods and use of this plasmid. Please note that Addgene's sequencing results identified discrepancies when compared to the assembled sequence provided by the depositing laboratory. The first GFP construct in this plasmid (bp#441-1154) is yeast codon optimized and contains a E124K mutation and amino acid 206 is A (not K, as suggested by the depositor's sequence). The second GFP construct in this plasmid (bp# 1212-1928) is NOT yeast codon optimized. This construct has successfully been used by the depositing laboratory as detailed in the associated publication and the sequencing discrepancies are not a concern for the function of the plasmid. RRID:Addgene_35199 Chloramphenicol and Ampicillin PMID:22848718 Generation of ϕC31 transgenic Drosophila for GAL4 driven RNA inhibition (RNAi). 2 inverted gateway cloning cassettes are flanked by gypsy insulator sequences to allow uniform transgene expression between insertion sites. 10 UAS binding sites, hsp70 basal promoter. ftz intron facilitates RNA hairpin formation and expression. Transgene selection using white gene. loxP site facilitates elimination of transgene markers via Cre recombinasemediated excision with ZH-attP landing sites. Ampicillin resistant. Chloramphenicol resistant for amplification. Please note that the inverted sequences present can cause rearrangements of this vector. Recommend miniprep only and confirm plasmid map subsequent to amplification or cloning. After gateway recombination, recommend amplification in Stbl2 cells [Invitrogen Cat. 10268-019] and combining minipreps for Drosophila injection. Addgene's sequencing results indicated a single nucleotide mismatch at bp# 5286 when compared to the full plasmid sequence provided by the depositing laboratory. This difference does not affect plasmid function. Please see the associated publication for more information: Wang J-W, Beck ES, McCabe BD (2012) A Modular Toolset for Recombination Transgenesis and Neurogenetic Analysis of Drosophila. RRID:Addgene_35196 GLT1 POL1 24xPP7SL Synthetic Ampicillin and Kanamycin PMID:21512033 Note that Addgene NGS results found 22xPP7 stem loop repeats rather than expected 24. It is not known how this impacts plasmid function. This plasmid was designed to construct mRNAs that contain multiple copies of an RNA sequence (the 24xPP7 stem-loops) that can be recognized by a chimeric RNA-binding protein fused to GFP, encoded in plasmid pDZ276 (Addgene plasmid #35194). See associated publication for detailed methods and use of this plasmid. Please note that the LacZa-ccdB fusion in this plasmid is not functional and was originally present in the vector backbone to aid in screening for insert. Can't find your Plasmid?

We recommend that you click next to the search bar to check some helpful tips on searches and refine your search firstly. If you want to find a specific plasmid, it's easier to enter an RRID or an Addgene Catalog Number to search. You can refine the search results using Facets on the left side of the search results page. If you are on the table view, you can also search in a specific column by clicking the column title and enter the keywords.

If you still could not find your plasmid in the search results, please help us by registering it into the system — it's easy. Register it with Addgene.
