Vector Backbone Description: Backbone Marker:Scott Sternson; Vector Backbone:AAV2; Vector Types:Mammalian Expression, AAV, Cre/Lox; Bacterial Resistance:Ampicillin
Defining Citation: PMID:30111097
Comments: Sternson Lab recommendations for propagation of AAV-FLEX plasmids: "These vectors are prone to recombination. This is a well known issue with these AAV vectors and is due to the inverted terminal repeats (ITRs) required for AAV production. To minimize recombination, we propagate these plasmids in NEB Stable cells. Also, to minimize recombination, cells should be cultured at 30 C.
Note that these cultures will grow slowly (20 h for minipreps). Better yields and culture times are obtained with 2xYT as the media. This is strongly recommended.
Because recombination may still happen occasionally, we do a panel of restriction digestions to assess whether the ITRs are in tact. Separate digestions with PvuII, Sma1, and SnaB1 should be performed."

Species: E. coli
Genetic Insert: gRNA
Vector Backbone Description: Vector Backbone:psgRNA; Vector Types:; Bacterial Resistance:Chloramphenicol
Defining Citation: PMID:29765036
Proper citation: RRID:Addgene_114006 Copy
Species: E. coli
Genetic Insert: colicin E1 immunity protein
Vector Backbone Description: Backbone Marker:Novagen/EMD Millipore; Backbone Size:5227; Vector Backbone:pET52-b; Vector Types:Bacterial Expression; Bacterial Resistance:Ampicillin
Defining Citation: PMID:27795317
Comments: This protein was never expressed independently. It was cloned in order to subsequently clone a colicin gene, which would be lethal in the absence of the immunity protein. The cloned fragment was from base pair 500 to base pair 48 of ColE1, which encompasses the E1 immunity protein promoter and the gene.

Can't find your Plasmid?
We recommend that you click next to the search bar to check some helpful tips on searches and refine your search firstly. If you want to find a specific plasmid, it's easier to enter an RRID or an Addgene Catalog Number to search. You can refine the search results using Facets on the left side of the search results page. If you are on the table view, you can also search in a specific column by clicking the column title and enter the keywords.
If you still could not find your plasmid in the search results, please help us by registering it into the system — it's easy. Register it with Addgene.
