Species: Xenorhabdus nematophila ATCC 19061
Genetic Insert: NrdJ-1C - XtpS
Vector Backbone Description: Vector Backbone:pCDF; Vector Types:Bacterial Expression; Bacterial Resistance:Spectinomycin and Streptomycin
Comments: This plasmid encodes the termination unit of the non-ribosomal peptide synthetase (NRPS) called xenotetrapeptide synthetase (XtpS), split at an XUTII site and fused to the C-terminal part of the split intein NrdJ-1.When co-expressed with pIN092 and pIN201, the three fragments splice into a functional NRPS that produces the non-ribosomal peptide (NRP) called xenotetrapeptide (XTP; cyclo[Leu-D-Val-Val-D-Val]; CC(C)C[C@H]1C(=O)N[C@@H](C(=O)N[C@H](C(=O)N[C@@H](C(=O)N1)C(C)C)C(C)C)C(C)C; 410.6 g/mol).
For successful NRP production, the three plasmids must be co-transformed into an E. coli strain that expresses a broad-spectrum phosphopantetheinyl transferase, such as MtaA (DH10B::mtaA) or Sfp (BAP1), which is required for the post-translational activation of the T domains.
Expression is controlled by an araBAD promoter, inducible with 0.2% arabinose.

Species: Xenorhabdus nematophila ATCC 19061
Genetic Insert: SspGyrBC - XtpS
Vector Backbone Description: Vector Backbone:pCDF; Vector Types:Bacterial Expression; Bacterial Resistance:Spectinomycin and Streptomycin
Comments: This plasmid encodes the termination unit of the non-ribosomal peptide synthetase (NRPS) called xenotetrapeptide synthetase (XtpS), split at an XU site and fused to the C-terminal part of the split intein SspGyrB.When co-expressed with pIN103 and pIN203, the three fragments splice into a functional NRPS that produces the non-ribosomal peptide (NRP) called xenotetrapeptide (XTP; cyclo[Leu-D-Val-Val-D-Val]; CC(C)C[C@H]1C(=O)N[C@@H](C(=O)N[C@H](C(=O)N[C@@H](C(=O)N1)C(C)C)C(C)C)C(C)C; 410.6 g/mol).

Species: Arabidopsis thaliana
Genetic Insert: Telomere
Vector Backbone Description: Backbone Size:9189; Vector Backbone:pTF101.1; Vector Types:Plant Expression; Bacterial Resistance:Spectinomycin and Streptomycin
Defining Citation: PMID:17085598
Comments: Please note that Addgene NGS found a 1.5kb deletion in the telomere repeats as compared to the original depositor sequence. The depositor has confirmed that the telomere sequence in the plasmid is inherently unstable, and that the plasmid is still functional for telomere truncation.
The plasmid was also verified by restriction digest. The gel image is provided on this page under- Resource Information- Addgene Notes.

Species: Synthetic
Genetic Insert: CLOMELEON (CFP-YFP fusion with ENLYFQG-linker)
Vector Backbone Description: Backbone Marker:Andrew Gleave; Backbone Size:11612; Vector Backbone:pART27; Vector Types:Plant Expression; Bacterial Resistance:Spectinomycin and Streptomycin
Defining Citation: PMID:15086798
Comments: pART27::CLOMELEON is a binary pART27 vector harbouring a NotI-fragment with a gene cassette subcloned from pART7::CLOMELEON to stably insert the CLOMELEON gene (Kuner, T. & Augustine, G.J.; PubMed: 11055428; doi: 10.1016/S0896-6273(00)00056-8) into the genome of plants by Agrobacterium-mediated gene transfer, e.g. by floral dip (Clough & Bent 1998; PubMed: 10069079; doi: 10.1007/BF00028910). Can't find your Plasmid?
We recommend that you click next to the search bar to check some helpful tips on searches and refine your search firstly. If you want to find a specific plasmid, it's easier to enter an RRID or an Addgene Catalog Number to search. You can refine the search results using Facets on the left side of the search results page. If you are on the table view, you can also search in a specific column by clicking the column title and enter the keywords.
If you still could not find your plasmid in the search results, please help us by registering it into the system — it's easy. Register it with Addgene.
