  • RRID:Addgene_71505

http://www.addgene.org/71505

Vector Backbone Description: Backbone Marker:Promega; Vector Backbone:pGL3-Promoter; Vector Types:Insect Expression, flySTARR-seq screening vector; Bacterial Resistance:Chloramphenicol and Ampicillin
Defining Citation: PMID:25517091
Comments: This plasmid was made by replacing the sequence of pGL3-Promoter between BglII and FseI with a fragment containing the core promoter listed above, an mhc16 intron, an ORF (sgGFP, Qbiogene, Inc), a ccdB suicide gene flanked by homology arms (used for cloning the enhancer candidates during library generation), followed by the pGL3's SV40 late polyA-signal.

Vector Backbone Description: Backbone Marker:Promega; Vector Backbone:pGL3-Promoter; Vector Types:Insect Expression, flySTARR-seq validation vector; Bacterial Resistance:Chloramphenicol and Ampicillin
Defining Citation: PMID:23328393
Comments: We replaced the SV40 promotor of the pGL3-promotor plasmid by DSCP and inserted a Gateway-cassette into the multiple-cloning-site (MCS; using KpnI and BglII), i.e. upstream of the DSCP, to allow Gateway cloning of candidate sequences

  • RRID:Addgene_71509

    This resource has 10+ mentions.

http://www.addgene.org/71509

Vector Backbone Description: Backbone Marker:Promega; Vector Backbone:pGL4.10; Vector Types:Mammalian Expression, humanSTARR-seq screening vector; Bacterial Resistance:Chloramphenicol and Ampicillin
Defining Citation: PMID:23328393
Comments: Note: The original human STARR-seq plasmid has been used in the Arnold et al. 2013 publication. It should be replaced for all screens in mammalian cells with the updated version reported by Muerdter et al. 2017 for much improved signal-to-noise, especially for screens with highly complex libraries (e.g. those covering an entire mammalian genome). The improved version is Addgene plasmid # 99296. For enhancer-activity measurements in mammalian cells, please also consider the possibility of a plasmid-induced innate immune response (see Muerdter et al., 2017). The sequence between SacI and AfeI on pGL4 was replaced by the fragment containing the Super Core Promoter 1 (SCP1), a synthetic intron (pIRESpuro3, Clontech; cat. no. 631619) an ORF (sgGFP, Qbiogene, Inc), a ccdB suicide gene flanked by homology arms, and the pGL3's SV40 late polyA-signal.

Species: Gallus gallus
Genetic Insert: actin prom-dsRed-scramble shRNA
Vector Backbone Description: Backbone Size:3000; Vector Backbone:pUC; Vector Types:Mammalian Expression, AAV, RNAi; Bacterial Resistance:Chloramphenicol and Ampicillin
Defining Citation: PMID:25706476
Comments: To express shRNAs, we used the pPRIME system. This generates micro-RNA-30 (mir30)-derived shRNAs and co-expression of a marker protein, such as dsRED, from the same transcript. The shRNA hairpin sequences were placed into the mir30 site of pPRIME-cmv-dsRed-FF3 and then cloned into the AAV vector (Zhang et al 2015 Nat Neurosci), SbfI(or PstI) + PacI will retrieve the 2.1kb insert (dsRed-scramble shRNA-chloramphenicol) and the 5.1kb AAV-CBA backbone, which is ampicillin resistant. The SbfI site in the middle of dsRed has been silenced. The construct can be linearised with XhoI.

Species: Gallus gallus
Genetic Insert: dsRed and shRNA to knock down the mouse adra2 receptor
Vector Backbone Description: Backbone Size:3000; Vector Backbone:pUC; Vector Types:Mammalian Expression, AAV; Bacterial Resistance:Chloramphenicol and Ampicillin
Defining Citation: PMID:25706476
Comments: The shRNA hairpin sequences were placed into the mir30 site of pPRIME-cmv-dsRed-FF3 (Addgene Plasmid 11664; gift from S. Elledge, Howard Hughes Medical Institute). Inserts in pPRIME were released by SbfI and PacI digestion, and cloned into the PstI and PacI sites in the polylinker of the AAV-genome plasmid pAM-flex (Murray AJ et al.2011 Nat Neurosci 14, p297), to give ITR-cmv-Penhancer/chicken β-actin-dsRED-mir30-shRNA-woodchuck post-translational regulatory sequence (WPRE)-bovine growth hormone polyadenylation signal (pA)-ITR. The sh RNA knocks down mouse adra2a expression.

Defining Citation: PMID:25915022
Comments: Note: Addgene's quality control sequencing identified a Y64H amino acid residue substitution in the Hygromycin resistance cassette. This residue change is not thought to affect the plasmids function or ability to confer resistance to Hygromycin.
