Genetic Insert: BSD
Vector Backbone Description: Backbone Marker:Alfred L. Fisher; Vector Backbone:pMOD4-galK-G; Vector Types:Bacterial Expression, Mouse Targeting, FLP / FRT; Bacterial Resistance:Ampicillin
Comments: PCR amplify around the 5' T2A and 3' FRT site, making sure to add a PacI site at the 3' of the FRT site.
As suggested in the plasmid name, due to a PCR error, the rpsL open reading frame has a 1-bp deletion that results in a frame shift and as a consequence premature stop codons. Despite this, the downstream BSD open reading frame is translated from the bicistronic RNA in E. coli without problems. These mean that the mutated rpsL cannot be used for streptomycin counterselection, but the unmutated BSD can and was successfully used several times to confer blasticidin resistance for selection after recombineering.

Comments: Four copies of miR-124 target sites (Åkerblom et al., 2012, https://doi.org/10.1523/JNEUROSCI.0558-12.2012) were cloned into “StLa” reporter plasmid (pCAG-loxP-pEM7-Neo*-bGHpA-3xSV40pA-loxP-tauLacZ-bGHpA from Nam and Benezra, 2009, https://doi.org/10.1016/j.stem.2009.08.017) without the tauLacZ insert and the Rosa26 homology arms. The 4x miR-124 target sequence can be released with BamHI and NotI.

Can't find your Plasmid?
We recommend that you click next to the search bar to check some helpful tips on searches and refine your search firstly. If you want to find a specific plasmid, it's easier to enter an RRID or an Addgene Catalog Number to search. You can refine the search results using Facets on the left side of the search results page. If you are on the table view, you can also search in a specific column by clicking the column title and enter the keywords.
If you still could not find your plasmid in the search results, please help us by registering it into the system — it's easy. Register it with Addgene.
