Comments: Rat IRS-3 was generated by PCR from the rat genomic DNA sequence. The intron was removed by PCR overlap extension using the following two pairs of primers: 5'-CGTGGATCCGCGATGAAGCCTGCAGGTACG-3' (sense) plus 5'-CTTGGGGGCTGAAACCCATGTTTGCTGGGCA-3' (antisense) and 5'-TGCCCAGCAAACATGGGTTTCAGCCCCCAAG-3' (sense) plus 5'-GCTGTCGACGTTCTAGAACTTGATGCTG-3' (antisense). BamHI and SalI sites were introduced at the 5' and 3' ends, respectively, of the coding sequence of IRS-3 by PCR. The subsequently generated coding sequence of rat IRS-3 was ligated into BamHI and SalI sites in pBABE-puro.
