Neuronal calmodulin levels are controlled by CAMTA transcription factors.

Thanh Thi Vuong-Brender | Sean Flynn | Yvonne Vallis | Mario de Bono
eLife | 2021

The ubiquitous Ca2+ sensor calmodulin (CaM) binds and regulates many proteins, including ion channels, CaM kinases, and calcineurin, according to Ca2+-CaM levels. What regulates neuronal CaM levels, is, however, unclear. CaM-binding transcription activators (CAMTAs) are ancient proteins expressed broadly in nervous systems and whose loss confers pleiotropic behavioral defects in flies, mice, and humans. Using Caenorhabditis elegans and Drosophila, we show that CAMTAs control neuronal CaM levels. The behavioral and neuronal Ca2+ signaling defects in mutants lacking camt-1, the sole C. elegans CAMTA, can be rescued by supplementing neuronal CaM. CAMT-1 binds multiple sites in the CaM promoter and deleting these sites phenocopies camt-1. Our data suggest CAMTAs mediate a conserved and general mechanism that controls neuronal CaM levels, thereby regulating Ca2+ signaling, physiology, and behavior.

ChIP-seq (tool)

RRID:SCR_001237

Set of software modules for performing common ChIP-seq data analysis tasks across the whole genome, including positional correlation analysis, peak detection, and genome partitioning into signal-rich and signal-poor regions. The tools are designed to be simple, fast and highly modular. Each program carries out a well defined data processing procedure that can potentially fit into a pipeline framework. ChIP-Seq is also freely available on a Web interface.

Calmodulin Target Database (tool)

RRID:SCR_008443

This database contains nearly 300 sequences known to bind calmodulin, published as of March 2003. Calmodulin is an intracellular calcium receptor found ubiquitously in eukaryotes. It is capable of regulating biological activities of many cellular proteins and transmembrane ion transporters mainly in a Ca2 -dependent manner. When the intracellular calcium level rises to 10-5 M, four Ca2 ions bind to calmodulin, and this Ca2 -calmodulin complex binds the target proteins, initiating various signalling cascades. Calmodulin has four EF-hand motifs that change conformation upon binding calcium ions. Each EF-hand motif contains two alpha helices connected by a 12-residue loop. The calcium ion binds to the loop region and changes the relative positions of the alpha helices (Yap et al, 1999). In absence of calcium, the alpha-helices in the EF-hand motif of calmodulin are positioned almost parallel to each other. This is known as the closed conformation.
